I did my Diploma Thesis in Prof. Dr. Michael Przybylski's group for Analytical Chemistry at the University Konstanz, Faculty of Chemistry between September 1995 and June 1996 (under direct care of Privatdozent Dr.Michael O. Glocker). The title was:

Selective modification of cysteine thiol groups with derivates of arsonous acid for the characterization of disulfide structures in proteins and folding intermediates of the Macrophage-Colony Stimulating Factor (M-CSF).

Synthesis und characterization (NMR, UV, MS) of two arsonous acid derivatives (Melarsen Oxide).

UV-spectroscopic analysis of the chemical selectivity and the time course of the modification reaction with dithiols in presence of the thiol-free reducing agent tris-(2-carboxyethyl)phosphine (TCEP).

Reaction of the synthesized arsonous acid derivatives with modell peptides (oxytocin, vasopressin) / proteins (BPTI, insulin) and mass spectrometric characterization of the products.

Characterization of the stability of the arsonous acid dithio esters.

Carrying out the refolding reaction for the cytokine Makrophage-Colony Stimulating Factor (M-CSF).

Stabilization of the M-CSF folding intermediates by chemical trapping with the synthesized melarsen oxide and with iodoacetamide.

Determination of the molecular weight and modification degree of the modified M-CSF beta folding intermediates by mass spectrometric and SDS-gelelectrophoretic methods.
